由yl9193永利杨宝学教授主编的《Aquaporins》一书已由Springer出版社正式出版发行。来自中国、美国、日本等11个国家的水通道研究领域的专家学者参与了撰写。该书系统介绍了近30年来水通道蛋白(Aquaporins,AQP)领域的研究进展。全书共分18章,其内容包括水通道的分子生物学、水通道的进化过程、水通道转运特性、水通道与腺体分泌、神经系统的水通道、心血管系统的水通道、呼吸系统的水通道、消化系统的水通道、泌尿系统的水通道、生殖系统的水通道、皮肤的水通道、眼的水通道、胚胎发育与水通道、尿崩症、水通道和肥胖、水通道作为治疗靶点、具有水通透性的其他膜通道和水通透性测定方法等。
水通道蛋白是一组特异性通透水的膜通道蛋白。该家族共有13个成员(AQP0~AQP12),包括单纯通透水的水通道和通透水及甘油等小分子的水甘油通道。水通道广泛表达于肾脏、红细胞、脑、眼、心脏、血管、肺、肝脏、肠道、膀胱、输尿管、睾丸、腺体等多个组织器官。水通道基因敲除小鼠模型研究和临床流行病学调查发现水通道功能缺陷可导致尿崩症、干眼症、干皮症等病理改变,研究还发现水通道功能性抑制可减缓脑水肿和肿瘤的进展,提示水通道蛋白可作为新的药物靶点,其抑制剂有可能研发成为新型治疗脑水肿和肿瘤的药物。以水通道蛋白为靶点的药物发现近年来正逐步成为该领域的研发热点,水通道的生理学功能研究、药物靶点确认和相关药物筛选模型在不断发展之中。

· The mechanisms and physiological functions of water transport across biological membranes are subjects of long-standing interest. Recent advances in the molecular biology and physiology of water transport have yielded new insights into how and why water moves across cell membranes. Aquaporins (AQPs) are a group of water channel proteins that are specifically permeable to water and some other small molecules, such as glycerol, urea, etc. Thirteen water channel proteins (AQP0 – AQP12) have been cloned, and gene organization, protein crystal structure, expression localization and physiological functions of some AQPs have been studied and determined. In recent years, the studies in aquaporin knockout mouse models suggest that aquaporins may be involved in some disease development and be useful targets for drug discovery of selective inhibitors. Our aim in writing this book is to stimulate further research in new directions by providing novel provocative insights into further mechanisms and physiological significance of water and some small molecule transport in mammals.
· This book provides a state-of-the-art report on what has been learned recently about aquaporins and where the field is going. Although some older work is cited, the main focus of this book is on advances made over the past 30 years on the biophysics, genetics, protein structure, molecular biology, physiology, pathophysiology and pharmacology of aquaporins in mammalian cell membranes. It is likely that advances in understanding molecular biology and physiology of aquaporins will yield new insights into biology and medicine.
